Í þessu verkefni er markmiðið að rannsaka hlutverk umritunarþáttanna Rhox5, Rhox6 og Rhox9, sem tilheyra Rhox fjölskyldu homeobox umritunarþátta, í ákvörðun frumkímfruma í músum.
Frumkímfrumur eru forverafrumur kímfruma. Ef sérhæfing eða þroskun frumkímfruma fer úrskeiðis leiðir það til ófrjósemi. Þess vegna er brýnt að skilja mörkun, sérhæfingu og þroska kímlínunnar. Í þessu verkefni höfum við afhjúpað nýtt hlutverk fyrir Rhox umritunarþætti í þessum ferlum og greint í smáatriðum bindingu þeirra og net umritunarstjórnar snemma í fósturþroska. Við höfum jafnframt framkvæmt svipgerðargreiningu með því að nota CRISPR-Cas9 og ræktir stofnfruma úr fósturvísum músa. Þessar niðurstöður hafa eflt skilning okkar á kímlínunni og munu jafnframt verða grundvöllur þekkingar okkar á frjóvgun og eðlilegum fósturþroska.

This project aims to investigate the role of the Rhox family homeobox transcription factors Rhox5, Rhox6 and Rhox9 in the transcriptional control of mouse primordial germ cell specification. Pirmordial germ cells are the founder cells of the germ line and the failure in their specification will give rise to sterile animals. It is therefore imperative to understand the specification and development of the germ line. In this project we have revealed a novel role for Rhox transcription factors in this process and analyzed in detail their binding and transcriptional control network in early embryonic lineages. We have furthermore performed a phenotype analysis using CRISPR-Cas9 and embryonic stem cell cultures. These results have enhanced our understanding of the germ line and will further underpin our knowledge of the processes underlying fertilization and normal embryonic development.
